Narrative:
Received a reroute from clearance delivery after pushback from the gate. Controller read the full route phonetically; starting with pumkn as the first fix. We inserted it into the [flight computer] as was read to us. We did not notice that the [flight route] started with the first fix being pmpkn. After departure were cleared direct pmpkn; and when the aircraft started the turn; the departure controller immediately gave us a vector to heading 060 and queried us about the spelling of the fix.

Title: A320 Captain reported a track deviation when they navigated to PUMKN instead of PMPKN after takeoff from ORD.
Narrative: Received a reroute from clearance delivery after pushback from the gate. Controller read the full route phonetically; starting with PUMKN as the first fix.
