Narrative:

As the tower said; 'line up and wait runway 1R;' we informed the tower the cabin was not ready for take-off. The tower replied; 'advise; hold; advise ready for take-off; line up and wait 1R and advise ready for take-off.' the first officer read back and continued running the checklist. As we were moving the first officer had doubt whether the tower meant to hold short and advise or to line up and wait 1R and advise ready. He confirmed with the tower [we were to] line up and wait 1R. The tower confirmed and replied; 'thank you.' we pushed off the gate late due to late arrival from previous leg; inflight was not ready; the pre departure clearance had an amendment that ground control re-amended on taxi out and changed the clearance back to original filed; but most importantly the tower instructions were vague. Each one of us understood something different and the workload was high. Clarify more and verbalize what each one of us understood; especially in light of the multiple changes taking place.
